MCW Protein Facility Silver Stain Protocol

Adapted from Blum et al, Electrophoresis, vol. 8, pp 93-99, 1987, and Shevchenko et al, Analytical Chemistry, vol. 68, pp 850-858, 1996

45% methanol, 7% acetic acid	fix	30 min
milli-Q water	wash	3 x 5 min
Na2S2O3.5H2O 0.2 g/L (freshly made)	incubate	90 sec
milli-Q water	wash	3 x 30 sec
AgNO3 0.2 g/100 ml	incubate	25 min minimum
milli-Q water	wash	3 x 60 sec
Na2CO3 6g/100ml, 37% formaldehyde, 50µl/100ml, Na2S2O3.5H2O (from step 3) 2ml/100ml	develop	until stained
45% methanol 7% acetic acid	stop	10 min
